(ISF) Spam Filtering Methodology

I am looking for advice and/or comments on which is the better method (and why) of blocking spam/viruses/malware etc. As far as I can tell there are three methodoligies for this:

1) Appliance Based (eg. Barracuda, etc.)

2) MX Redirection (eg. Mx Logic, etc.)

3) Internal Software (eg. Spamtitan, MacAfee, etc)

We are a fair sized organization with 600 email users and we receive approximately 15000 external emails a day.
