Short answer: No.

If you want to add messages to an already defined domain,
you can only
do that in i18n folders. That's the reason all the domains
used by Plone
itself are still in i18n folders.
If you want to overwrite translations the only bullet-proof
way is to
put them inside an i18n folder directly inside your
$INSTANCE_HOME,
right next to bin, products, Extensions, ...
Overwriting messages inside a domain specified in a locales
folder is
currently not possible. If you really need it, you could
look into the
CustomizableTranslations product in the collective, whose
functionality
is going to be added to Plone at some point.
