If you know how to edit styles from the 'Edit' menu click
'Output
Styles'. When you see 'Citation' Template - simply change
the the
brackets you want.  Then in the 'Bibliography' Template you
can make
changes to the journal styles accordingly (Italics, bold or
whatever).
It's a bit tricky in the beginning, but once you know what
changes you
are making and see them happen, you will have better control
over it.

The Vancouver style in my EndNote (X.0.2) uses square
brackets but not
italicized journal/book names. It seems there are
variations. The one
from UQ at

"In the Vancouver Style, citations within the text of
your essay/paper
are identified by Arabic numbers in round brackets. This
applies to
references in text, tables and figures. e.g. (2) - this is
the style

None of the variations that I found uses italicized
journal/book names.
